      选取单瓣长寿花‘萨姆巴’的叶片为外植体,设计正交试验,研究不同种类植物生长激素及其浓度配比的培养基对长寿花叶片愈伤组织的诱导、不定芽的分化和不定芽生根的影响,并研究试管苗炼苗及移栽的成活情况。试验结果表明,诱导长寿花叶片产生愈伤组织的最适培养基是MS+6-BA 1.0 mg/L+NAA 0.2 mg/L+2,4-D 0.2 mg/L,诱导率是83.33%。诱导愈伤组织不定芽分化的最适培养基是MS+6-BA 2.0 mg/L+NAA 0.3 mg/L,增殖系数是21.4。最适的不定芽生根培养基为1/2MS+ IBA 0.2 mg/L + NAA 0.2 mg/L,生根系数为7.03。试管苗移栽到蛭石∶营养土=1∶1的基质中培养,移栽的成活率可达100%。

      The leaves of single-flowered Kalanchoe blossfeldiana were chosen as explants, and orthogonal experiments were conducted to investigate the effects of various proportions of different plant growth hormones on the callus induction of leaves, adventitious buds differentiation of callus, root regeneration of seedlings. The transplanting of seedlings was also explored in this study. The results indicated that the optimal medium of K. blossfeldiana callus induction was MS+6-BA 1.0 mg/L+NAA 0.2 mg/L+2,4-D 0.2 mg/L, and the callus induction rate was 83.33%. The optimal medium of adventitious bud differentiation from callus was determined to be MS+6-BA 2.0 mg/L+ NAA 0.3 mg/L, and the propagation coefficient was 21.4. For the optimal root regeneration of adventitious buds, the medium was 1/2MS+0.2 mg/L +0.2 mg/L, with a rooting coefficient of 7.03. The survival rate of plantlets reached 100% in the meteorites: nutrition soil (1:1) matrix.
